Can a pollyester layup be made over rustseal? If so what is the procedure?

Yes, you'd need to prep the surface and apply a couple of coats of RustSeal. Let that dry approx. 12+ hrs, then scuff the area with an 80 grit, blow off any dust, then proceed with your layup.
